Abstract : Introduction: The aim of this Ph.D. dissertation was the search for antiplasmodial and Gholamreza Asghari, Seyed Esmaeil Sadat Ebrahimi, Jerzy W. Jaroszewski, Javad Asiliantimicrobial agents from Platycladus orientalis as a native coniferous plant in Iran. This plant was chosen because of moderately antimicrobial and antiplasmodial activities obtained in the preliminary screening of essential oils of fruits and ethyl acetate extracts of leaves and branches respectively.Experimental: The essential oils from different parts (leaves, branches, fruits) of P. orientalis were obtained by steam distillation (SD) and steam distillation solvent extraction (SDE) methods separately. The essential oils were analyzed for identification of the components by GC and GC-MS instruments. Also the antimicrobial compound using biaoutography method was isolated.Several pure diterpenoides were isolated from ethyl acetate extracts of leaves and branches using VLC, CC, TLC, normal and reverse phase HPLC. Their structures determined using 1D and 2D NMR methods, ion cycloteron HRMS, and optical rotation data. The compounds were tested for their in vitro antiplasmodial activity.Results: The main components of all essential oils were determined as .a-pinene, 8-3- carene, sabinene and cedrol. Significant differences between the two methods were. recorded in the oxygenated constituents percentages. The SDE showed a higher recovery of these compounds. Terpinen-4-ol was isolated as antimicrobial compound.Six labdanes (pinusolide, pinusolidic acid, isocupressic acid, lambertianic acid, 16-oxo-8(17),13-labdadiene-15,19-dioic acid and 15-hydroxy pinusolidic acid) and four isopimarane diterpenoids [8 (9),15- isopimaradien-313-ol ; 8(14 ),15 -isopimaradi en-3 (3,19-diol: 313.19-dihvdroxy-8(9),15-isopimaradien-7-one as new natural products and 15-isopimaren-3[3,813-diol] were isolated and their structures were elucidated using different spectroscopic methods. All tested compounds exhibited weak (IC50 > 25p.M) in vitro antiplasmodial effects.Discussion: The results showed that using the SDE method the recovery of oxygenated compounds is higher. Terpinen-4-ol an antimicrobial oxygenated compound that could be found in higher amount in fruits and also in essential oils obtained by SDE method.Some changes were seen in erythrocyte membrane shape, at concentration regions where the antiplasmodial activity were observed, indicative of their incorporation into the lipid bilayer and therefore the antiplasmodial effects of these compounds thus appear to be an indirect effect on the erythrocyte host cell, rather than a toxic effect on the parasites themselves..
