Abstract : Introduction: Everydays millions of people use different species of Allium as food or supplements. In this study, A. elburzense (bulbs), A. hirtifolium (bulbs and flowers), A. atroviolaceum (bulbs) and A. eriophyllum (bulbs) are phytochemically investigated for first time. The study resulted to separation and elucidation of several new steroidal saponins.Methods: The plant materials were air dried, powdered and extracted with hexane, CHC13, CHC13ت‍MeOH (9:1), and Me0H. The Me0H extracts were partitioned between butanol and water phases. The CHC13ت‍Me0H (9:1) and butanolic extracts were chromatographed by CC or MPLC on a R1318 Silica gel, using a linear gradient solvent system from H20 to Me0H as mobile phases. Interested fractions with saponins contents were selected, base on preliminary NMR analysis. The selected fractions were purified by preparative HPLC with the suitable mobile phases (mixture of Me0Hت‍H20). 'H NMR, 13C NMR, 2D NMR, FABMS and IR, techniques were used to elucidate the isolated compounds.Results: Phytochemical analysis of bulbs of A. elburzense leading to the isolation of thirteen furostanol and spirostanol saponins, eight of them are new, named elburzensosides Al/A2, B1/B2, C 1/C2 and Dl/D2. Study of A. hirtifolium flowers has lead to the isolation of high amounts of six new furostanol and spirostanol saponins, named hirtifoliosides A 1 /A2, B, C 1/C2 and D along with three known spirostanol saponins. High concentration of the three known flavonol glycosides have been also found both from flowers and bulbs of A. hirtifolium. Saponins or sapogenins were absent in the bulbs of A. eriophyllum and A. atroviolaceum.Conclusion: According to high concentrations of new isolated saponins on these plants, it is suggested that further pharmacological and structure activity relationship (SAR) studies of isolated compound should be done. In addition, study on other Allium species growing in Iran is recommended.
