Introduction: Hepatocyte growth factor/scatter factor (HGF/SF) is known to be a paracrine factor that is produced by stromal and mesenchymal cells and promotes the growth and scattering of various cells. The receptor for hepatocyte growth factor is a transmembrane tyrosine kinase. HGF/SF-Met signaling has been strongly implicated in the promotion of the invasive/metastatic tumor phenotype. Therefore we intend to measure the level of HGF and C-MET in lymph nodes of NHL patients and find a possible correlation with their stage as a marker of invasion. -Materials and methods: This is a descriptive-analytic study. 57 DLBCL patients with available Paraffin-embedded tissue samples enter the study. All the clinical and paraclinic data were gathered in a specific form. The stage of the patients at diagnosis was determined. HGF and C-MET protein expression was detected using the immunohistochemistry assay and the expression of both proteins was compared between differet stages. The percentages of positive cells were averaged to yield an immunohistological score of 0-100 . The following categories were defined: negative if positively stained tumour cells were below 10 and onepositve if between 10 to 50 and two-positive if more than 50 of cells were stained.Results: we detected the expression of HGF and C-met in tissue samples of DLBCL patients. 35 out of 57 (61.4 ) were negative for HGF and the rest expressed HGF. 20 patients expressed one-positive HGF and two patients expressed two-positive HGF. 23 patients (40.4 ) were negative for C-met and the rest exressed C-met.28 patients (49.1 ) expressed one-positive C-met and 6 cases (10.5 ) expressed two-positive C-met. The mean HGF expression in stagel was 21.44 and 30.03 for stage2 and also 35.47 for stage3 and 24.88 for stage4 (p=.063). Also the mean expression of C-met expression was 27.94 for stagel and 28.47 for stage2 and 30.50 for stage3 and 28.47 for stage4 (p=0.96). Conclusion: Our results indicate that both HGF and C-met protein is expressed in DLBCL patients. . Also the expression of both proteins increase from stagel to satge3 and in can be one of the mechanisms of cell spread and cell dissemination in lymphoma patints. However because of the limited number of cases it is not statistically significant. Also our study shows that the early stage is not accompanied by high expression of the protein. On the other hand the patients with satgel and high expression of the protein may need further evaluation for_staging or may need chemotherapy and rigid follow up..
