The adult mammalian brain contains endogenous precursor cells that maintain homeostasis and can also replace lost cortical and subcortical neurons following injury. The majority of investigations into factors regulating the activation of neural precursor cells have focused on the rostral lateral ventricles, despite evidence demonstrating the existence of precursor cells outside of this region. Given the paucity of information regarding the distribution of precursor cells, we employed three distinct assays to compile a detailed atlas of the number and frequency of precursor cells along the ventricular neuraxis of the adult mouse brain. The neurosphere assay, neural colony forming cell assay (N-CFCA), and label retaining cell approach each detected precursor cells throughout the ventricular neuraxis, totaling 3730▒276, 4275▒124, and 5292▒102 or 1186▒188 (5-month versus 7-month chase) respectively. However, the number of precursors detected in individual 400pm sections varied from a minimum of 8 to a maximum of 891 depending upon the rostral-caudal coordinate assayed, with the greatest variation between sections occurring in the rostral ventricular region typically assayed. In addition, by micro-dissecting the periventricular region from 6 individual 400i_tm sections we generated approximately 2700 primary neurospheres, as compared to the gross dissection of the same region, which (based on published work) typically produces 150 to 900 primary neurospheres. These results serve to explain the variation in precursor cell frequency previously reported, and highlight the need for caution in the employment and interpretation of results when using these assays for the investigation of resident neural precursors..
