Abstract : Background The p53 as a main tumor suppressor gene plays important roles in genomic stability. A common G-to-C polymorphism at codon 72 in the p53 gene affects on gene function and has been associated with increased risk of many cancers. This may be a marker for genetic susceptibility to breast cancer. It is believed that this polymorphism has different ethnic distribution. Microsatellite instability is considered a marker of genetic instability and seems to be important for the development of various human cancers. We aimed to investigate the microsatellite instability in Isfahanian breast cancer specimens with different p53 codon 72 genotypes.Methods We undertook a caseتcontrol study on 96 controls and 96 patients with breast invasive ductal carcinoma from the city of Isfahan. P53 codon 72 genotypes were identified using allele-specific PCR. The status of BAT26 marker microsatellite was evaluated using screening method of single-strand conformational polymorphism and silver staining. The x2-test was used to assess the significance of differences in the prevalence of p53 codon 72 polymorphisms and microsatellite instability, the P value was set at Ps0.05. Results:In control samples, the genotype distribution for p53 polymorphism showed 36.5 , 45.8 and 17.7 for the Arg/Arg, Arg/Pro and Pro/Pro genotypes, respectively. In the cancer group, 70.8 of the cases were Arg/Arg, 21.9 were Arg/Pro and 7.3 were Pro/Pro. Alteration of BAT-26 size was seen in 15 out of 96 (18.5 ) of our specimens. The differences in the distribution of the p53 codon 72 polymorphism between the cases and controls were statistically significant (P<0.001), but the distribution of Microsatellite Instability in different p53 codon 72 genotypes were not significant (P>0.05).Conclusion Findings of the present study indicated that p53 codon 72 polymorphism probably is a genetic predisposing factor for breast invasive ductal carcinoma development in Isfahanian specimens. Our data revealed that BAT26 microsatellite instability was not associated with risk of breast cancer in different genotypes of p53 codon 72. These results suggest that p53 codon 72 polymorphisms could be considered as a molecular marker, contributing to the genetic profile in breast cancer patient in Isfahan.Key words: P53 codon 72; BAT26 marker; Breast invasive ductal carcinoma; Microsatellite instability; P53 genotypes..
