Introduction: Superoxide radicals and oxidized low-density lipoproteins (oxLDL) has been proved to play a role in decreased NO-dependent vasodilator function in hypercholesterolemia and atherosclerosis. The 8-isoprostaglandin F2a is one of the most valid markers of in-vivo oxidative stress. We aimed to investigate the preventive and treatment effect of dietary supplementation of L-arginine (3 in drinking water) by measuring 8-isoprostaglandin F2a as a marker of oxidative stress. Materials and Methods: Rabbits were randomized to four experimental groups (n= 6*4): Control, Prevention, Treatment and Prevention & Treatment. All of them received hypercholesterolemic diet for the first four weeks. The Prevention group received supplementary L-arginine in the first four weeks, the Treatment group received L-arginine in the second four weeks and the Prevention & Treatment group received L-arginine throughout the experiment. The serum level of lipids and lipoproteins and fatty streaks were measured during the experiment. To determine the antiatherosclerotic effects of L-arginine the 8-isoprostaglandin F2a levels were measured using the ELISA method in the Results: No significant differences were observed for the serum levels of lipids and lipoproteins between the groups during the experiment. The serum concentration of 8-isoprostaglandin F2a was significantly decreased in the Prevention group rather than the Control group at the end of the experiment. The IMT ratio in the Prevention group was significantly lower than the Control group at the end of the experiment. Conclusion: In fact there is a preventive role for L-arginine in our hypercholesterolemic models.
