introduction: Postmenopausal hormone replacement therapy (HRT) is known to affect various aspects of lipoprotein metabolism. The estrogen receptor p (ER13) mediates the action of estrogen on the lipids and lipoproteins metabolism. The aim of the present study was to investigate whether the ERI3 A 1730G polymorphism modifies ABCA1 gene expression and metabolic responses to HRT in postmenopausal women.Materials and Methods: The population included 60 normoIipidemic postmenopausal women with the diffewit genotypes (20-GG. 20 GA. and 20 AA) assigned to 90-day hormone replacement therapy. The ERB genotype. ABCA I gene expression. oxidative status and lipid profile were determined respectively. by PCR-RFLP analysis, real-time PCR, FRAP. and ELI SA methods.Results: Analysis of leukocytes ABCA I rriRNA showed a significant increase in ABCA! gene expression after the postmenopausal HRT (P--0.0011). After the postmenopausal HRT. subjects with GG genotype had greater increase in ARCA I (P=0.002) than subject carrying A allele. Postmenopausal HRT led to significant increase in TAC (15 . P=0.02). There was no significant relationship between oxLDL/B2GPI complexes. TAC levels and leukocytes ABCA I gene expression even after adjustment for confounding factors.Conclusion: The beneficial cardiovascular effects of postmenopausal HRT could beexplained, at least in part, by increasing the ABCAI gene expression and improving.
