Herbal medicine as a result of multi-function, multi-target characteristics have potential of optimum pharmaceuticals and nutraceuticals effects on Ad patients. R.damascena as a natural plant with high source of flavonoids may have curable effect on treatment of AD. according to a narration of holy prophet Mohammad, peace be upon him and his pure family, "favorable odors enhance memory" we desigend to investigate the potential therapeutic effect of the R.damascena extact on learning and memory impairment in a rat model of AD via stimulation of adult neurogenesis and synaptic plasticity mechanisms. following extract preparation and HPLC analysis and toxicity studies, median lethal dose (LD50) and dose levels were determined. initially all animals were randomly divided in 6 groups and for evaluation of baselin training performance in behavioural tests, spatioal acquisition phase of Morris water maze test and training trialof passive avoidance task were performed . for induction of rat model of Alzheimer`s disease, Amyloid-B was injected into CA1 area of the hippocampus bilaterally using stereotaxic surgery. 21 days after injection, the first probetrials of the behavioural tests were done, and following the impairment of the learning and memory, for anlysis of the Amyloid-B plaques in the hippocampus, Congo red staining was performed. then R.damascena extract was administered orally at doses of 300, 600 and 1200 mg/kg/day via a gastric tube in gourps 4 to 6 for one month. sham and 3rd groups received only normal saline at the same time but control group didn`t receive anything. all animals received intraperitoneally injections of BrdU 50 mg/kg body weight twice daily in order to evaluation of proliferaion of the adult neural stem and progenitor cells and adult neurogenesis during the lat 7 days of R.damascena treatment. after that the secound probe trials of morris water maze test and passive avoidanc task were performed to evaluate the effects of extract on spatial and log term memories repectively. immunohistochemistry for BrdU and DCX was performed in order to evaluation of proliferation of neural stem and progenitor cells and migrating meuroblasts cells respectively. to analyze the synaptic plasticity and remodeling , ultrastructural study was performed using transmission electron microscopy. finally unbiased stereological study to measure the total voume of the hippocampus and absolute volume of its subfields was used. our resault showed that R.damascena significantly increases proliferation of the neural stem and progenitor cells and migrationg neuroblasts in the hippocampus in a dose-dependent manner. administration of R.damascena extract induced adult neurogenesis and synaptic plasticity mechanisms in a dose dependent manner and finally reversed the AB-associated memory abnormlities in a rat model of amyloid-B induced Alzheimer`s disease.
