Abstract

Introduction: Common variable immunodeficiency (CVID) is a heterogeneous group of the primary immunodeficiency disorders and the commonest primary immunodeficiency syndrome. CVID is accompanied by the hypogammaglobulinemia and recurrent bacterial infections This thesis aimed to determine the immunophenotyping of the peripheral blood transitional B cells and the B cell activation factor of the TNF family receptor (BAFFR) mutations in the patients with common variable Immune deficiency (CVID) with the allergic or non-allergic symptoms compared to the healthy controls.Methods: Genomic DNA was extracted from the whole blood of the patients with CVID and healthy controls (n=15) using an extraction kit (genet bio). Primers were designed to both sides of each exon. After confirming the accuracy of the PCR, the product was sent to the sequencing. The peripheral blood mononuclear cells (PBMC) were isolated by the Ficoll-Hypaque density centrifugation. For the transitional B-cell phenotype, samples were washed in phosphate-buffered saline before staining. The CD19-FITC (Exebio), CD24-PE (Exebio), and CD38 PE (BD Exebio) antibodies were used.The staining process was performed according to the manufacturers’ protocol and then the cells were run on a FACS Caliber, BD. The data was analyzed using Cell Quest software.Results: The results of the sequence and alignment of the sequences, did not show mutations in the gene .The flow cytometric indicated that the mean percentages of B cells in the allergic patients group (3.68 ± 0.96, p<0.001) and non-allergic patients group (1.2 ± 0.57, p <0.001) were significantly different from the controls (10.43 ± 3.46).
