Abstract	:	Objective: Many studies have supported the role of Protein Kinase C (PKC) inhibitors in the physiopathology and treatment of bipolar disorder in adults. Tamoxifen is one of the drugs with the effect of protein kinase C inhibitions. This study aims to determine the effect of tamoxifen on the rate of improvement mania symptoms in the sample of children and adolescents with acute mania.Materials and Methods: In this randomized, placebo-controlled clinical trial study, 44 patients with bipolar disorder with acute manic episode were gradually and randomly put in two intervention-treatment groups of Lithium (with the serum level of 0.8 -1.1) and tamoxifen (20-40mg per day) (22 subjects) and also Lithium and placebo (22 subjects). The main comparisons of measurement included the questionnaire of Young Mania Rating Scale (YMRS) and Children Depression Inventory (CDI) each of which was completed each week. The pharmacological side effects of serum level of lithium were examined weekly. ANCOVA test was used for the statistical analysis.Results: There was no difference in the base score of YMRS and CDI in both groups but there was a significant difference in these scores between and within the groups (p<0.05). Conclusion: The addition of tamoxifen to lithium causes a significant difference between two groups in terms of the effect on decreasing the symptoms of mania and depression and the rate of improvement compared to placebo.
