Abstract

Objective: Multiple sclerosis (MS) is a chronic autoimmune disease of the central nervous system (CNS) identified by axonal degeneration.Various agents including genetic and environmental factors are involved in the etiology of MS. The Osteopontin (OPN) and its receptor (Integrin α4β1: ITGA4) have important role in immune response, so their encoding gene could be considered as a candidate for susceptibility to MS diseases. In the present study, we evaluated the OPN serum levels, the frequency of OPN (rs28357094T>G) and ITGA4 (rs1449263A>G) polymorphisms in RRMS patients compared to healthy controls in Isfahan population. Methods: Blood samples were collected from patients with multiple sclerosis and healthy controls (n=100). DNA were extracted from whole blood (Genet Bio, Korea), and then OPN (rs28357094T>G) and ITGA4 (rs1449263A>G) polymorphisms were detected with HRM Real Time – PCR technique (Solis Bio Dyne co., Estonia). The serum level of OPN was measured by ELISA method (Biospes Co., China).The results were analyzed by SPSS 16.0 software package.Results: The frequency of TT genotype (OPN- rs28357094 T>G) (P.value =0.02, OR= 1.85) and AA genotype (ITGA4- rs1449263A>G) (P.value =0.012, OR= 2.1) were significantly high in MS Patients than the control groups. The mean serum level of OPN was significantly highr in the MS patients than the controls (P.value=0.03).Conclusion: Overall, the present study showed an important role for OPN in the pathogenesis of MS. Moreover, OPN-rs28357094 T>G and ITGA4-rs1449263A>G polymorphisms could have associated with susceptibility to multiple sclerosis disease
